An antibody specific for S100A16 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyS100A16 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for S100A16 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of S100A16 bound in the initial step. The color development is stopped and the intensity of the color is measured. There are at least 21 different types of S100 proteins. The name is derived from the fact that the protein is 100% Soluble in ammonium sµLfate at neutral pH.Most S100 proteins are homodimeric, consisting of two identical polypeptides held together by non-covalent bonds. Althoµgh S100 proteins are structurally similar to calmodµLin, they differ in that they are cell-specific, expressed in particµLar cells at different levels depending on environmental factors.
